Question:

A Question on Electrophoresis?

How does the rate of migration(ROM) of DNA during electrophoresis correlate to the voltage applied and size of the DNA fragment used?

Meaning, how ROM correlates to VOLTAGE.

And, how ROM correlates to DNA FRAGMENT SIZE.

And, I need to know of some experiments to prove this point.

And, the amount of Agarose Gel used. (0.5%, 1% etc.)

Thanks, in advance.

  1. A higher voltage gives a faster ROM.

A larger DNA fragment size gives a slower ROM (larger fragments are slowed more by the gel).

A more concentrated gel has a smaller pore-size, and therefore gives a lower ROM.
