  1. if the gene of interest if tagged with amp resistance, then you can plate the e.coli on plates (after diluting) with agar and amp, and the ones that grow have the gene of interest. then counting the colonies that grow and multiplying by the plating factor and dilution factor will tell you the origional concentration of the gene of interest.

    if the numbers seem wrong, remember that spontaneous reversion or contamination could have occured, altering your count.
