I take it you are talking about Eleftherios Venizelos airport? Cape Sounion is 69 km southeast of Athens. EV Airport is 30 km northeast of central Athens by road. You may be able to hail a taxi from the airport but it will be VERY expensive (upwards of at least 100 Euros!). Your best bet would probably be to take the bus to Athens for the night (they leave every half hour from the airport) and then take a bus or taxi down to Cape Sounion in the morning.
