  1. Streak plate method requires more skill comparatively, coz it finally depends on this technique that whether u get an isolated colony or lawn of growth.

I find streak plates to be pretty easy for the most part, as you usually do get clear separation of colonies by the third streak.  When you refer to slants n broth, I assume you are talking about using them to mix two or more samples.  In that case, I find that harder, as I find working with broth annoying, especially waiting for it to dry.  Just my own opinion.

  3. Streak plate because there is a knack to not letting the different sections overlap while streaking and then while it is culturing. This gives you much clearer definition of the growth.

  4. I don't have a lot of experience with this sort of thing (only one semester so far), but I think the streak plate method takes the most skill. When plating a culture that has several different types of bugs in it, it can be quite a challenge to get a good isolate of each kind.
