1). Each microorganism requires  specific Nutrient media basing on that you have to select essential nutrients,along with required distilled water.

2). If you want solidified media you have to add agar other wise don't add agar.

3). Sterilize the media using autoclave for required time.

4). After cooling to room tempareture you have to innoculate the microorganism,if it is broth.If it is solidified media you have to streek.

5).Incubate for specified time.

6).Sterile conditions are very very important for good culture of microorganisms.
