Yes you can, there are many components that will allow you to incorporate a shopping cart, just make sure you get the latest version of Joomla (1.5.6) as there is security issues with the legacy versions

Joomla runs on the mysql database. You can install Virtuemart shopping cart for ecommerce or iDigistore works well also
