There is one in Errol Street Nth Melbourne.

Never my scene, there's enough to laugh at and about in Melbourne without stage. But you will find one there and a neat old part of the city too.

I recall the Comfortable Chair bar on Lygon St, Brunswick East, used to have open mic comedy nights on a Mon/Tues.  That one that might be worth checking out.
