  1. Perform a steak plate of the bacteria. The purpose is to isolate single colonies. Each colony represents one type of bacteria. Based on the appearance of the colonies you should be able to differentiate between the two types of bacteria present.


  2. Bacterial isolation can be done by the streaking technique. A sterile loop is used to pick up some bacteria from a small colony, and the material is streaked on one quarter of the plate. The loop is flamed, and some material is picked up from the end of the last streaking, and is spread another quarter of the plate. This process is then repeated until the whole plate is used. Single colonies will show up.

  3. ummm...not sure how totally right this is but MacConkey agar is selective and differential correct? It grows Gram negative and will produce color for the lactose fermenting. Then you pick from colonies that are clearly different (diff color, size, etc) and then streak each one one diff plates or slants...no???
