  1. Spec of the bikes, for one.  

    Superbikes are generally based on 1000cc production machinery, with limits on what can be modified.  Don't recall the exact rules, but I believe that you have to use the original engine cases, etc.  There are also minimum weight requirements.  

    MotoGP bikes are prototype machines.  They aren't based on any bikes in the manufacturers' lineup.  They have minimum weight requirements, a maximum amount of fuel they can carry, and displacement limits, but that's about it. The bikes are worth millions, and riders are paid in the millions.  Valentino Rossi makes about $30 million per year, making him one of the highest-paid athletes anywhere.

    MotoGP is regarded as the most prestigious.  You'll find a lot of riders going from MotoGP to SBK as their career winds down, (Max Biaggi), or from SBK to MotoGP as they're successful (Colin Edwards).
