The ORI is the specific DNA sequence where replication begins.  The RNA primer binds to the ORI to initiate the process.

There are no amino acids that are coded for by the stop codons so no amino acids are added to a polypeptide chain from any of the stop codons.

The 40s and 60s rRNA subunits bind together to form the eukaryotic ribosome.  From there the mRNA binds to the initiation site and translation begins.
