I am supposed to grow my bacteria in minimal media and use for making a microbial fuel cell. The only problem is that i don know how much do i inoculate in my 25% minimal media from a broth culture? what does "use 25% minimal media" mean? does it mean to use 250ml of originally prepared 1L media?

  1. yep to inoculate you just take a few colonies from the agar plate and put it in your 25% minimal media.  Usually we use a tooth pick to scrape a few colonies, and drop it in, you dont need many.

The 'use 25% minimal media' is exactly how you said.  Dilute the minimal media to 25%.  So if you wanted 1L total, 750mL of distalled water and 250mL of minimal media.  Be careful to check its not already diluted for you, often it is.
