Does coagulation need heat to occur?

i just wanna know if coagulation of milk needs heat to occur providing that it has an enzyme and cofactor or can it happen without heat.

Heat is the transfer of thermal energy, not the existence of thermal energy.  So to say that something "has heat" at room temperature is kind of meaningless unless you are talking about a changing temperature (in which case you are talking about heating or cooling, not just having heat).  Sure, adding heat to milk can cause it to coagulate.  But add lemon juice without adding heat and it will coagulate.  There you have coagulation without adding heat.  The chemical reaction requires thermal energy to proceed, though--i.e. it would not happen at absolute zero, but then nothing happens at absolute zero, so that is meaningless too.
