Why are the cells fragile after the addition of Polyethylene glycol (PEG)?

  1. Hi Angelz,

    The properties of Polyethylene glycol (PEG) that make it useful in promoting cell membrane fusion, are also responsible for causing cell fragility.  

    PEGs mechanism of action is volume-exclusion aggregation of membranes and dehydration of areas in contact.  The ability of PEG to alter the distribution of water molecules on the surface and interior of the membrane can induce 'rigidation' of the membrane, making it fragile. Combined with its capacity to induce aggregation of phosphatidylcholine (a main membrane phospholipid), PEG can greatly weaken the integrity of the membrane.
