  1. The best way to get to the Cinque Terre from La Spezia is to take the train. You can buy the Cinque Terre pass at the train station in La Spezia for 8 euro and that allows you unlimited train rides between La Spezia and Sestre Levante, with stops in all five of the 5Terre and access to the walking trail between the towns.

    If you have your heart set on a boat ride, I'd recommend doing it between the towns. However if you have your heart set on a ferry from La Spezia, here is a link that will give you all the information you need about taking the boat in that area.

  2. I think taking the ferry along the Cinque Terre is the best thing to do. The view is amazing. Looking at the little villages perched in hill tops is quite a sight. I would take a taxi from La Spezia to Potrovenere and then catch the ferry from there along the Cinque Terre to the furthes town (Monterosso), hop off the ferry and then start walking along the trail through the rest of the towns, back to Riomaggiore, then catch the ferry back to Portovenere and then the taxi back to La Spezia. You could conceivably do it in one day, I did, but you have to walk pretty fast and it is better to do it slower. I went back again and spent one day at each village, which was a lot better. You could also not do the walk between the villages and just explore each village and then catch the boat to the next. The train is very convenient, but the boat is so much nicer.
