From Penn Station you have three options each day:

1.  The Maple Leaf Train 63 departs Penn Station at 715 AM arrives Buffalo at 329 PM, 7 days a week.

2.  Empire Service Train 281 departs at 1020 AM arrive Buffalo at 655 PM, 7 days a week.

3A.  Empire Service Train 283 departs at 120 PM arrive Buffalo at 935 PM, Sunday through Thursday.

3B.  Empire Service Train 285 departs at 220 PM arrives Buffalo at 1035 PM, Friday and Saturday.

The earlier you plan your trip the cheaper tickets are.  The Maple Leaf sells out very early and is currently sold out into early August.

Take Amtrak train number 49.  It goes up to Albany, adds a section from Boston, and continues to Buffalo and ends up in Chicago.
