First off you may want to go into the services by Start > Run > services.msc / and disable Remote Registry, and Routing and Remote Access.  Get Comodo Firewall Pro (free) and install it, that should give you control over your PC again. http://www.personalfirewall.comodo.com/

Disconnect your internet connection, save backup of all your precious files on externel disk, reformat your harddisk, after that reinstall antivirus first with latest update, and walla! you are completely clean, no trojan, no rootkit,..

It always work, rather than scanning 250 GB Harddisk, it will take forever to finish..
