Given that this enzyme degrades phenol, back titration against a known number of equivalents of phenol added to the phenolase in solution is probably one path to an assay for the original phenolase enzyme concentration.  One could also use gas-chromatography to quantitate the phenol in the solution before introduction of the phenolase and after introduction of the phenolase as phenol is a semi-volatile.  Here one would need to use an extraction technique perhaps solid phase extraction or liquid-liquid.  One might also be able to use an antigen indicator for phenol concentrations.  This does assume that the phenol degradation is strictly due to phenolase.  There are other paths to degradation for phenol....UV, bacteria,adsorption, etc.  Matrix effect and extraction losses as well as interferences and instrument response problems have been well characterized for GC-MS analysis of phenol in a variety of initial matrix substrates by the EPA as this is an environmental contaminant targeted in cleanup studies.
