you need to go out and buy a wireless pci card for your desktop and if your router is wireless and online you will have no trouble having them both online at the same time

To clarify what exactly do you want to do:

1.  Use your laptop wireless connection with your desktop?  (ie using the laptop as your wireless router)--Not going to happen unless your laptop is a server.

2.  Using your wireless card (AT&T, Verizon, ClearWire, etc) in your desktop -- nope again.

4.  Setting your desktop up with a wireless network (ie. you have a DSL or Cable connection with a wireless router and you want to have your desktop wireless)--> Yes this can be done after you purchase a wireless NIC card or USB device.
