How can we have colonies that are only bacterial or fungal?

  1. We streak a sample over the surface of an agar medium. Each subsequent streaking is like a further dilution of the sample. Eventually we get colonies each from a single bacterium or a fungal spore. The culture can be further purified by restreaking from a colony.
