How do I email about a job post on Craig's list?

I would simply like to know what I would need to say, any questions I should ask. If the job post does not specify time deadlines or job requirements, should I straight out ask about these things in my first contact with the person? Thanks.

Anything that was not covered in their ad on craigslist that I wanted to know I would not hesitate to ask in my first email.
