  1. Okay - since it is quite a distance away you will want to take the night train. This saves you time for sightseeing as you travel during the night, refreshed (?) in the morning to start your tour of Lisboa! There is a 1045pm departure from MAdrid Chamartin station that arrives into Lisbon at 813am the following day. Check out  http://www.raileurope.com/us/index.htm

If you are doing quite a bit of traveling in Europe you might want to consider a rail pass. They have a Eurail Spain & Portugal pass that might be good if you will be using the train at least 3x. Check it out http://www.raileurope.com/us/rail/passes...

Now, on your train trip, don't forget to take lots of bottled water, some good snacks and wear a comfortable pair of clothes!

  2. There is a daily night direct train connecting Madrid and Lisbon. Below is the link to the timtable from Renfe, Spanish train company.

It's a 'trenhotel' train, a train with a wide range of confort options: from a normal seat (88 euros return) to a high class single room (305 euros return: has private shower, toilet, phone, dinner and breakfast included...)

Maybe a supplement is applicable if you use a european train pass.

  3. Sorry, but there´s no train that goes from Madrid to Lisbon.
