  1. transport against the concentration gradient has to be by active transport

    active transport is either primary atthe expense of ATP or secondary by using the energy of another gradient

There are two ways things can enter/leave the cell, actively or passively.  Passive transport is usually performed along a concentration gradient, e.g diffusion where ions move from high  concentration to low concentration; whilst active transport requires ATP inorder to facilitate the transportation and is often against a concentration gradient.  An example could be sodium pumps where ATP is used to actively pump sodium out of cells.  The Na concentration is normally higher outside cells so it diffuses into the cell, creating an unfavourable environment.  Sodium pumps therefore pump the sodium back out of the cell against the concentration gradient.

  3. active transport. When the substance encounters the cell the cell membrane encapsulates it and takes it to the center of the cell.
