Start and stop codons are on the mRNA strand.  The ribosome scans along the mRNA, looking for the start codon, AUG.  When AUG is in the P-site of the ribosome, a tRNA carries methionine in to begin the process of translation.  Translation continues until a stop codon (there are three to choose from) enters the A site.  When this happens, there is no matching tRNA and the ribosome disassembles and the polypeptide is cleaved from the final tRNA.
