The main problem is that Ancona is on the other side of Appennini mountaigns, starting from Florence.

I recommend you the fastest way is you take the A1 Highway from Florence to Bologna, then switch to A16 Highway from Bologna to Ancona (you don't have to get out and in the highways... they are chained, just do follow the indications).  

This will result in a 328Km trip (about 204 U.S. Miles) and an average 4 hours of car driving, including a little stop in the mid of the trip to have some brief rest.
