It depends on a few factors. Traffic can be a absolutely hellish at times! It also depends on where in the Hamptons. Westhampton is closer to to the City than Southampton or East Hampton. Also, anything further east of Southampton and you're left to local roads with a limited speed limit.

If you left on a Saturday morning, early and before the traffic, it should take about two hours to get to Southampton. Less to Westhampton and more to Bridgehampton or East Hampton.

There are a few routes, too. The quickest during low traffic would be the Grand Central Parkway/Northern State to the Sagtikos Parkway, south to Sunrise Highway. Sunrise east right into Southampton. There are plenty of other options, some more scenic than others.
