  1. Probably forever. Penicillin is a poor choice for skin infections, though you could get lucky.

There's a reason you aren't supposed to take antibiotics unless they're prescribed by a professional. Your grandmother doesn't have the expertise to recommend an appropriate drug.

  2. generally any antibiotic taken orally takes 72 hours to start seeing relief. however penicillin isn't the best choice for what you describe . also not wise to take someone else's antibiotics or any drugs older than a year as some turn toxic in the system causing even more unwanted problems with one's health .

  3. I think you need to see a doctor.  "Forever" may have been a pessimistic answer, but it's probably the right one.

Something filled with pus may need to be drained before any antibiotics are effective.

Penicillin would be a useless choice against a Staph infection, which is one possible reason for what you describe - let your doctor figure out what the infection actually is and prescribe an appropriate drug.

Penicillin has to be taken several times a day, not once.

Old medicine from your grandmother may not be safe or effective even if it were taken at the right dose, at the right frequency, for the right kind of infection.  One of the most common reasons for drugs not working or doing harm is someone getting old medicine from their grandmother or uncle or sister or mother.
