I'm assuming you are talking about in the US.  You don't apply, you are either nominated or you work your way up through the state dept.   Check out the state dept website for more info.

If you're not in diplomatic service already, you've got no chance because an ambassador's position  is the topmost rung on a very long ladder

that job is appointed by the President of the United States,.....if you are not friends with the President then you have no chance,.....
