What is the best way to promote an online service in order to build a regionally focused user-base? What methods work best to convert real people into online users?

You can target PPC advertising by city, state or region so that the only users are in your targeted area.  Of course you can also target websites for banner ads in this same way.  Naturally, if you can build an email list of prospects in your geographic area, you can market to them that way.

Most mail firms that allow you to advertise to their list can target by geography as well.

The best way to convert is dependent upon your product, the sales process and the generic or custom nature of your sale.  If you have experience selling in person, you just need to customize the sales process online to replicate the experience as much as possible.
