  1. The  coarse adjustment k**b is the large k**b that is used to focus your specimen first.  Place your specimen on  the stage using the proper procedure in producing a correctly mounted specimen.  Look through the eye piece and turn the k**b until you get a clear picture in the eye piece.  Secure the slide with the stage clips so it does not move.  Now change the objectives from low to medium (if you have 3 objectives) and refocus the slide, but use the fine focus not the coarse focus.  If you use the coarse focus you may crack the slide and damage the lens.  Now turn to high power and refocus using the fine focus again.  Never use the coarse focus on medium or high power since slide damage could occur.

  2. Most microscopes have two main focusing k***s: The Coarse Focusing k**b and the Fine Focusing k**b.

The coarse focusing k**b is your heavy focus to get your image in approximate focus.  It's the larger of the two k***s.

The fine focusing k**b allows for minute movement in the focus, so you can get the exact detail you want.  For multiple layers on a slide, you can switch focus from one cell later to another with the fine focus by simply rotating it in the direction you want.

Other than that, just play with it a little.  Get a slide, put some cells on it (by slicing a cross section off a bit of celery, or swabbing the inside of your mouth with a cotton swab, put your glass cover plate over your sample, and look through!  Once you get a little practice in, it'll become second nature.

    Hope this helps! :)

  3. Fine adjustment k***s on the side of the microscope are used for bringing objects into focus when studying an object on a slide. On some microscopes, these k***s are not positioned concentrically as shown here.
