I'm having a hard time coming up with a good concept. I already have characters but i dont know what to write?

I have detailed out my characters but I have been reading different mangas and watching animes. I dont want to use their concepts but come up with a real good one. My friend suggested to use a orb that is a replica of earth and if its destroyed the world will be blown up. That to me sound a bit superhero saving earth i want some thing more fantasy. First of all, you should have thought up a story before the characters.  But that's ok. I'm personally inspired by the Bible.  I like to take a verse and think about what kind of neat story I could make by varying the plot a little and adding my own characters and flair.  I've also been thinking about writing a story about Cinderella, but from the stepsister's point of veiw.  you might not have any characters that would fit ino that category, but I thought that the idea was pretty cool.  You can use it if you want.
