I can't understand 1 point of dna sequencing via chemical method... ! plz help me !?

in chemical sequencing, we mark the 5' end with 32p........and at last we measure the size or length of dna fragments from this end........ bt dna molecule has two 5' end......so, from electrophoresis , we should get two same size dna fragments from two dna templete......then hw can we dertermine the sequence?

  1. for THAT method of sequencing you have to start with SINGLE-stranded DNA....that's in order to prevent the two signals you mention.
