Yes. Some banks might hold a check like that for about 10 days, just to make sure it is a good check. If you go online or walk into your bank they'll tell you if the funds are available, but 10 days for large checks is common.

If it hasn't there is definitely something wrong. Call your bank and ask them.  Jk

If it was a payroll check, a cashier's check, or issued by the state or federal government then it should have cleared the next day.

If it was a personal check then it would take 5 to 10 days to clear depending on the bank.

Best way to know for sure is to call the bank and ask, but three weeks is more than enough time.

Do you know the source of the check?  There are so many check scams that involve bogus checks.  Even if the bank releases the funds, a bogus check can take 2 months before being returned.

So it this check was from a lottery, or written for extra that you are suppose to wire to someone,  don't take the money out yet.
