Little Boy was a U-235 gun type bomb, they had done so many experiments with U-235 before the drop that it was never tested before being dropped on Hiroshima.

Because plutonium fissioned so rapidly, the Fat Man bomb design was tested at Trinity, it used a sophisticated implosion lens assembly of the subcritical shells of the plutonium core, and only then was it used in combat over Nagasaki.
