  2. For a 4" main drain you will have to use 2"by6"s which are really only 1.5" by 5.5" to have room for the closet bend and toilet flange.

  3. Not sure just how you think your moving your toilet, but let's assume you are talking about running the pipe above the floor to relocate the toilet. You could do this with a raised platform, assuming the height you have from the platform to the ceiling is still at least 6 foot. Your platform will have to be at least as high as the pipe you want to run, and accomodate the sweep elbows you would need to use to turn the pipe along the floor and then back up to the relocated toilet. You should also note that the additional pipe run will make this toilet more prone to clogging.

  4. not really sure I understand the need for a platform.  if you are moving the toilet across the room in the same 'bay' between floor joists, you would just extend the drain pipe, and find some way to get the water supply from the wall to the other wall or floor of new location.  you would have to rip up the subfloor between the 2 locations. if you are moving it to another joist bay, much bigger job: have to box out the joists to the other location. in either case, a platform would not serve much purpose.
