Yes.  It would.  However, it would change the weight by such a small amount to make it not worth it, unless it was a very large box filled almost exclusively by helium.

So yes, it would make the box lighter.  

No, it would not work practically.  You would spend far more than you save.

nope the hellium in the ballon will cost you more than the weight reduction it will provide.
