  1. There are various derivations to calculate the Vmax and Km from a michaelis menten curve.  

    Because the curve is V (y-axis) vs. [S] (x-axis)

    [S] is found at at the volume of 1/2 Vmax.  If that is not given you can simply replot the data now using 1/V and 1/[S] for y and x respectively to give you a linear line.

    The y intercept of this linear line is going to be 1/Vm and the x intercept will be -1/km  and the slope will be Km/Vm.

    This is kind of hard to explain in words, but if you just google a picture of a michaelis menten curve you will see what I'm talking about.
