Not really, but Helium displaces Oxygen in your lungs and when you breathe too much of it you can pass out or even asphyxiate.

Large amounts can cause asphyxiation, also some sources of helium gas can contain trace amounts of arsenic.  

Overall it's relatively harmless

Just put in in a balloon first, if you take it directly off the tank and make a mistake you can explode your chest cavity.
