Question:

Is it possible that microaerophiles can grow on agar surface keep in an air-tigth jar?

i am working on culturing microaerophiles from environmental sources. After first cultivation in air-tight jar, those microaerophiles were found at subsurface level, and so i subculture them on agar plate in air-tight jar as well..But tis time they grow faster and they appeared on surface of agar instead of subsurface..is this possible? or did i make any mistakes?

  1. Perhaps the culture observation has come out different because of lack of oxygen. When there was oxygen it appeared on the surface and due to lack of sufficient oxygen, the sub-surface case was observed.

    Microaerophiles prefer growing in an aerobic condition in presence of sufficient oxygen on agar surface.

    BUT, yes you can also grow it in a closed jar or bottle. as it needs lower concentration of air(02) to grow.

    But if it is grown in a vacuum environment, its growth would be faster in the beginning and then will slow down later.
