Is it possible to block someone from leaving messages on your 360?

Yes, go to your page and click on ignore this person, its on the left hand side below your avater, easy peasy.

If you mean IM messages, when you read them, you have an option. click on "ignore" and you will not hear from them again.

I assume you press the ignore button on their page.

There is an ignore this person underneath their selected picture.  Click that word and you will no longer recieve messages from them.
