Is there difference between the original Uncharted: Drake's fortune and the platinum version in terms of game-play or additional features?

No, they are exactly the same game. The only difference is that the game is now in the budget titles Platinum series (effectively just different packaging and a new price), and that it carries trophy support.

The original game was given trophy support via an update.

Actual gameplay wise, it is exactly the same.

Yes, the price.  But otherwise, the games are exactly the same.  No new content, no new game play.  Nothing.
