  1. the cell will need to be fixed first, otherwise you'll never get the antibody into the cell, and if you did, it would be degraded.  fixing the cell = killing the cell


  2. One has to get the antibody inside ..which means permeabilizing the cell..generally non-conducive to cell integrity. why not look for the mRNA transcripts of the protein

    ( in situ hybridization ) as one must alter the cell's membrane anyway. Autoradiography using radio sulfur and C14 can also be used..Also . Western blotting of cell contents using a fluorescent, colorimetric or chemiluminescent endpoit can be used to show the presence a cytoplasmic protein
