Jobs. Retail assistant, Assistant Manager, Manager?

I am 17, in year 12 but I'm looking for a job that offers about 4 or 5 days a week.

With the positions, how do you work your way up? From retail assistant to assistant manager?

Do you have to do a course or something?

Can you just apply for Assistant Manager jobs?

It all depends on your experience.  I assume because you are only 17 you haven't been working in retail for long if at all.  In which case you will have to start at the bottom, I would be very suprised if you would be offered an assistant manager job straight off.  Once you have some more experience you can either work your way up or you will have that experience on your CV in which case you can apply for the assistant manager roles elsewhere.
