Well, "best" and "most powerful" are not synonymous terms.  Some of the best man-stoppers are only mid-level in the power department.  The .357 magnum puts out a little under 600ftlb of energy, but is arguably the best defensive caliber on the planet.  More powerful calibers tend to over-penetrate, and expend most of thier energy in the busload of nuns sitting behind the bad guy, rather than in the bad guy himself.    

So, are you looking for the "best" or the "most powerful" caliber pistol?

Excluding rifle caliber pistols, the S&W .500 Magnum is the King.

Practically?  The .357 Magnum, .45 acp or 10mm.  The 10mm being the most powerful of the three.

i would say the desert eagle 50 cal. but you can buy pistols chambered for rifle ammo. these would be the most powerful

well the most powerful is probably the S&W .500 Magnum but why do you want power ? .......,well some  advice from me power only gives you Recoil,and weight(yes they build the gun tougher usually so the powerful cartridges wont damage the gun)  on youre gun the recoil makes it harder to hit anything with ,a good gun is a 9mm,.357 magnum,or a .38 special those are guns that are/were used by law enforcement and they are proven to be able to stop almost anyone ,if you hit them in the right places(chest,head) so that sums it up
