please tell me that why saline solution is used for staining and culture of microorganism?

microorganisms are cultured on media prepared with saline water so please tell me what is the work of saline solution

  3. Saline solution is 0.9 percent sodium chloride in water.

    Most living cells survive best in this concentration as this is the concentration that is equal to the inside of the cell.

    The purpose of the saline is to prevent the cell from drying out. However, if you use normal water, there is the possibility of bursting the microorganism because the cell is salty (and more concentrated) on the inside, so pure water from the outside will move into the cell to try and balance this, making the cell swell.

    If the solution is too salty, the opposite occurs, and the cell will shrink as water moves out to the more concentrated solution to try and balance it.

    That's why saline solution 0.9 percent is used to prevent any movement of water into or outwards, and keeps the micro-organism happy.

  4. if we want to culture a particular microorganism we want only that organism to be present in that ,when we open the lid of the culture dish there is possibilitie for the accumulation of other organism from the air ,to prevent this we use saline solution
