  1. Some larger post offices are open on Saturday.

  2. It depends what you're looking to do.

If you're just looking to use a Yucho Bank ATM then even some of the smaller offices might have ATMs open on Saturday/Sundays. But, you should try to get their before 5pm.

If you need other postal banking/insurance services then unfortunately you'll probably have to wait until Monday.

If you just want to buy stamps or mail a letter then most of the bigger branches will probably be open. Tokyo Central Post Office near Tokyo Station should be open to about 5pm or 6pm and they should have an after hours window as well.

Here's a map in case your not sure where it is.

In big city such as Tokyo some certain delivery offices are exceptionally open. (small non-delivery offices are closed on weekend and holidays)

One or two  delivery office opens on saturday  must be in at least each one Ward or City in Tokyo. They are out of service for banking facility but taking care of postal service only though.

  4. The central post office in each ward (for example the one on Miyamasuzaka near Shibuya station) is open on Saturday and also have 24-hour service counter.
