  1. well, if you're talking about spots it means you're talking about a 2D (2 dimensional gel). If so, each spot should represent a different protein, hence, the number of spots equals the number of proteins.

    Having said that, it is important to remember that: in the unlikely case of two protein with the same pI and mass - they will both occupy the same place and create only one spot. and, if you are using radioactive labelling than you won't see all the proteins, you will rather see only the newly synthesized proteins.
