Question about protein methods?

I have a bacteria that has on its membrane a protein(the protein is not a receptor).

I don't know the protein sequence and protein structure.

I also don't know which antibodies can binds with that protein.

How can i identify that protein using Western blot or Elisa.

Please write all that steps that should be done in order to identify that protein and explain them.

  1. 1)  Purify protein

    2)  Mix protein with adjuvant (complete Freunds, Incomplete Freund's or other of choice).  This helps you get better antibody titers.

    3)  Inject protein into an animal - usually a goat, sheep or rabbit.

    4)  Harvest serum and purify antibodies using affinity chromatography.

    5)  Test antibodies out in Western blot format.
