Will this affect my battery power?

My digital camera can record video. There are two sizes 320x240 and 640x480. I know the bigger size will take up more room on my memory card. Will the bigger size affect my battery? Will it make the battery power run out faster?

No effect on battery life. You're powering the whole memory whether it's empty, full or somewhere in between.
