Wikipedia says:

a restriction enzyme makes two incisions, once through each sugar-phosphate backbone (i.e. each strand) of the DNA double helix.

It says two incisions, but only tells of the one through each strand of the sugar-phosphate backbone.

  1. DNA is made up of two complimentary strands, each with its own sugar-phosphate backbone.  The restriction enzyme cuts through the sugar-phosphate backbone on each of those two strands = two total cuts.


  2. That is what it means, one incision on each of the two strands of DNA adds up to two total
