You did not provide the means of transportation (road/rail/air) you wish to use. So, let me try to give info for all three transports. This might help others in future with same query.

AIR:

Although, the closest airport is Nasik (100 Km) followed by Aurangabad (150 Km), I’m not aware of any direct flights from Chennai to Nasik or Aurangabad. You can fly to Mumbai and get a connection to Aurangabad. Alternatively, fly to Pune (Air Deccan DN-684 or SpiceJet SG-231) and then take another flight from Pune to Aurangabad (Jet Airways 9W-102) You can also hire taxi/bus to Shirdi from Mumbai or Pune.

RAIL is the best option if you want to use surface transport. You can take various routes.

Chennai – Bangalore City JN - (Train 2627 departs at 7:20 PM) – Kopargaon (reaches 1: 40 pm). Kopargaon is just 18 miles from Shirdi.

Chennai – (Train 2603 or 2759) – Secunderabad – (Train 7058) – Manmad. Manmad is 85 Km from Shirdi (Take Bus/Taxi)

Chennai - (Trains 2603 or 2759) - Hyderabad - (Train No. 349) – Aurangabad – (Bus/Taxi-150 Km) – Shirdi

Take a train to Pune JN from Chennai Central (Trains like Mumbai Mail, Mumbai Express, Chennai Express reach Pune at 11:20 PM, 9:35 AM, 2:25 PM respectively). From Pune, there are many trains to Kopargaon:

7305 (NZM LINK EXP), 2779 (GOA EXPRESS), 2129 (AZAD HIND EXP), 1601 (PUNE MMR PASS), 1333 (SHIRDI FAST PASS), 1321 (PUNE NZB PASS), 1077 (JHELUM EXPRESS), 1039 (MAHARASHTRA EXP)

Journey from Pune to Kopargaon is 4.5 to 5.5 hours depending on the train. You can also hire bus (AC/non-AC) or Taxi from Pune to Shirdi.

ROAD:

Distance is about 1300 Km. If you are planning to drive (quite a long distance to drive!!!), then your best route will be as below:

From Chennai take NH-4 until Palamaner-AP (195 Km) via Sriperumbudur, Narasingapuram, and Chittoor.

From Palamaner, take NH-219 towards Madanapalle (about 55 Km) via Punganur

From Madanapalle continue heading north on NH-205 until Anantapur (about 170Km from Madanapalle) via Kadiri

At Anantapur, merge on NH-7 and head towards Kurnool and go about 55 Km until Gooty.

From Gooty, turn right towards Bellary on NH-63 and reach Hitnal, KA (about 160 Km from Gooty) via Guntakal, Bellary and Hospet.

Then head north on NH-13 until Solapur (188 Km) via Bijapur.

From Solapur, take NH-211 until Bagpimpalgaon (220 Km) via Barshi, Bid and Georai.

At Bagpimpalgaon, turn left (west) towards Shrirampur and reach Shirdi via Shrirampur and Rahta Pimplas.
